NH65 starts at Pune, located in the western region of Maharashtra, and travels eastwards towards its endpoint in Machilipatnam, a coastal town situated on the Bay of Bengal. The highway passes through major cities such as Solapur, Hyderabad, Suryapet, Vijayawada and many others along its way.
The section between Hyderabad and Vijayawada is known as the Vijayawada-Hyderabad Expressway, a major expressway connecting the two cities. The highway also passes through picturesque regions such as Bhigwan, a small town in Maharashtra known for its bird-watching spots.
